Why test for Potassium?

Potassium is a critical factor in acid salt formation and tartrate precipitation. Many potassium additives are present in wine and beer for the following purposes: potassium carbonate reduces the levels of acidity; potassium metabisulphite is an anti-oxidant and preservative; and potassium sorbate is used to stabilise wine and prevent renewed fermentation. Excessive potassium levels contribute to higher pH measurements in wine and potentially tartrate instability problems. Testing for the optimised potassium concentration is vital to ensure product quality and taste.

Testing for Potassium

The following assay provides accurate determination of potassium levels in a wide variety of matrices. The test kit is applicable for both manual use or for automation on a range of analysers. The test protocol incorporates a specific enzymatic reaction followed by UV method of absorbance, the method is outlined below:

potassium wine and beer testing

Potassium is determined enzymatically via Potassium-dependant pyruvate kinase activity using phosphoenolpyruvate as a substrate. The pyruvate formed reacts with NADH in the presence of LDH to form Lactate and NAD. The corresponding decrease in absorbance at 340 nm is proportional to the potassium concentration. This simple yet highly specific test protocol ensures rapid results with minimal human error. This product is currently supplied to the wine/beverage and food producers globally.
